چکیده مقاله:

Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/m. The CR-39 is a special and sensitive plastic used to verify exact position of heavy-ions traversal. Terminal deoxynucleotidyl transferase (TdT) is an enzyme labeled with [3H] dATP for detection of cellular DNA damage by autoradiography assay. Results: The track of heavy ions traversals presented by pit size was almost similar for all different doses of radiation. No pits to show the track of traversal were found in 20% of the cell nuclei of the irradiation. Apparently these fractions of cells wave not hit by heavy ions. Conclusion: This study indicated the possible usefulness of both the CR-39 plastics and DNA labeling with TdT method for evaluating the biological effect of heavy-ions in comparison with low LET ionizing radiation.

Detection of terminal deoxynucleotidyl transferase.

The recently published study of peripheral blood lymphocyte subpopulations in patients with acute immunodeficiency syndrome, by Murray et al,’ may have contributed additional insight into normal T cell ontogenesis. However, the data on terminal transferase (TdT) must be interpreted with caution. It is stated that TdT levels are especially elevated in acute lymphocytic leukemia (ALL) of T cell o...

Terminal Deoxynucleotidyl Transferase (TdT) Inhibiti on of Cord Blood Derived B and T Cells Expansion

Purpose: Terminal deoxynucleotidyl transferase(TdT) is a DNA polymerase that is present in immature pre-B and pre-T cells. TdT inserts N-nucleotides to the V (D) J gene segment during rearrangements of genes, therefore, it plays a vital role in the development and variation of the immune system in vertebrates. Here we evaluated the relationship between cytokines like interleukin-2 (IL-2), inter...

Murine Terminal Deoxynucleotidyl Transferase

Terminal deoxynucleotidyl transferase (TdT) 1 is an enzyme which has the unique property of polymerizing deoxyribonucleotides onto a primer in the absence of a template (1-4) . Chang (4) first reported that TdT is present only in the thymus among various avian and mammalian tissues examined . Her finding was extended to humans by McCaffrey et al . (5), who found that the enzyme was restricted t...

Proton beam dosimetry by CR-39 track-etched detector

Background: High and intermediate energy protons are not able to form a track in a solid state nuclear track detector (SSNTD) directly. However, such tracks can be formed through secondary particles created during primary radiation nuclear reactions in a SSNTD. Materials and Methods: The protons with primary energies of 9.6 and 30 MeV available at the cyclotron accelerator with corresponding lo...

Terminal deoxynucleotidyl transferase (TdT), cytochemistry, and membrane receptors in adult acute leukemia.

Terminal deoxynucleotidyl transferase activity, immunologic membrane markers, and cytochemical reactivity were examined in blasts of 80 adults with acute leukemia. The study was interinstitutional and prospective so that some assessment could be made of the usefulness and practicality of these tests in the classification of adult acute leukemias. Initial classification as to the type of acute l...
